Introduction:

PAGE Gel Preparation Kit (fixed concentration) contains various reagents for PAGE gel preparation, including pre-mixed stacking gel solutions and separating gel solutions. Customers only need to prepare a small amount of pure water and prepare the required gel according to the instructions. Just mix the corresponding reagents in a 1:1 ratio before the gel is to be poured, no need to dilute and no need to overlay the separating gel with water, allowing quick casting of multiple gels. The upper gel (stacking gel) is colored to make the sample wells clear and easy to identify and load samples. The dyes do not affect electrophoresis and subsequent staining and membrane transfer experiments. This product adopts the Tris-glycine gel chemistries system and the gels can be used for both nondenaturing and denaturing electrophoresis. For the gels used combining with WSHT’s Ultrafast Running Buffer (Cat. #: EZB2006) and fluorescence loading buffer Quick Bands (Cat. #:ES008), the entire process of sample processing, electrophoresis, and photography can be completed within half an hour, and ideal electrophoresis results can be obtained.

1. The kit should be stored at 4°C in dark, and the coagulant should be stored at -20°C. The shelf life is one year.

2. If the coagulant is stored at 4°C, the shelf life is three months.

 

Protocol

1. Preparation of separating gel

Referring to the table below, take equal volumes of Separating Gel Solution and Separating Gel Buffer (reagent C and reagent D), mix well; add the corresponding amount of PAGE Gel Coagulant (reagent E) at a ratio of 1% last and mix well just before the gel is to be poured. Immediately after the separating gel solution has been prepared and mixed, fill gel cassette to approximately 2 cm from the top edge of the plate.

2. Preparation of stacking gel

1Referring to the table below, take equal volumes of Stacking Gel Solution and colorful Stacking Gel Buffer(reagent A and reagent B), mix well; then add the corresponding amount of PAGE Gel Coagulant (reagent E) at a ratio of 1% and mix well just before the gel is to be poured.   

2There is no need to cover the separating gel with water or wait for the separating gel to polymerize after pouring the separating gel. Immediately add the stacking gel on the separating gel. Be careful and gentle when pouring stacking gel to prevent the stacking gel from mixing with the separating gel. Fill gel cassette with the stacking gel solution and insert a comb without trapping air under the teeth. Wait for 20-30min to let it gelate.

3The gel coagulation time is related to the temperature. The higher the temperature, the shorter the time required for polymerization. The amount of PAGE Gel Coagulant used in the specification is under the standard condition of 25°C. Please increase or decrease the amount of coagulant according to the actual temperature. The maximum addition amount of PAGE Gel coagulant is 1%, the fastest polymerization time is about 15-30 minutes in summer, and about 30-60 minutes in winter with this ratio.

The specific number of gels that can be prepared is depend on its thickness and size.

Note:

  1. There will be errors in the volume of different gel making equipment, the above values are for reference only.
  2. Volumes of stacking gel and separating gel differ according to the thickness of gel casting.

3. Store the prepared gel

We recommend prepare PAGE gels fresh. If the prepared gel needs to be stored for a long time, please immerse it in the gel preservation solution (Cat. #:GP016), keep it sealed and moist, and place it in a cool dark place. Gels in preservation solution can be stored for 3 months at room temperature and 1 year at 4°C.For long-term storage of the gel, it should be noted that:

1) Avoid repeated violent changes in temperature.

2) Avoid the temperature below zero, the gel will freeze and be completely scrapped.

3) Make sure the gel is wet.

4.Electrophoresis

If traditional Tris-Gly running buffer is used at 180V, the electrophoresis will be complete in 60min. When the bromophenol blue indicator band is electrophoresed to the bottom of the gel plate or the predetermined position of the experiment, the electrophoresis can be ended. If using WSHT's Ultrafast Running Buffer (Cat.#:EZB2006), electrophoresis can be completed in as little as 20 minutes.

 

Notice:

  1. After taking the kit out of the refrigerator, please return to room temperature before use.
  2. Due to the special physical and chemical properties of dyes, it is normal that there is precipitation at the bottom of reagent B. Please shake well before use.
  3. The Separating Gel Solution and Stacking Gel Solution contain Acr and Acr-Bis, please be careful when handling, and pay attention to effective protection to avoid direct contact with the human body or inhalation.
  4. The dosage of PAGE Gel Coagulant is for reference only, and the actual dosage can be adjusted according to personal experimental habits and experience.
  5.  In the process of gel preparation, if it is necessary to further accelerate the gel polymerization, an appropriate amount of TEMED can be added as needed during gel preparation.
