Introduction:

Protein Sample Loading Buffer for Tricine is used to prepare protein samples for denaturing gel electrophoresis using tricine gels. The sample buffer is formulated with Coomassie Brilliant Blue as tracking dyes and DTT, SDS as denaturant. It is designed for optimal electrophoresis of small proteins and peptides using Tris-tricine polyacrylamide gels.

Storage: Store at -20°C for one year. It is recommended to freeze and store in aliquots to avoid repeated freezing and thawing.

Protocol (for reference only):

1. Mix Protein Sample Loading Buffer for Tricine (2×) with your protein sample with 1:1 ratio.
2. Heat the sample mixture in a 1× dilution at 100°C for 3-5 minutes to denature proteins completely.
3. Directly load the sample mixture into the wells of the PAGE gel after it has cooled to room temperature.
4. The electrophoresis can be stopped when bromophenol blue reaches near the bottom of the PAGE gel.

Notes:

1. This product is offered in 2× format and it should be diluted into 1× working solution according to the experimental requirements.
