Introduction:

The Three-color Prestained Protein Marker (11-180 kDa) is a three-color protein standard with 10 pre-stained proteins covering a wide range molecular weight for 11-180 kDa. Proteins are covalently coupled with a blue chromophore except for two reference bands (one green and one red band at 25kDa and 75kDa respectively) when separated on SDS-PAGE (Tris-glycine buffer). The Marker(11-180kDa) is designed for monitoring protein separated during SDS polyacrylamide gel electrophoresis, verification of Western transfer efficiency on membranes (PVDF, nylon or nitrocellulose) and for approximate sizing of proteins. The ladder is supplied in gel loading buffer and is ready to use format without heating and reductant. The concentration of each band is about 0.2-0.4 mg/mL.

Storage:

Store at -20℃ for one year.

Protocol:

  1. The protein ladder is supplied in a ready-to-use format for direct loading onto gels; no need to heat, reduce, or add sample buffer prior to use.
  2. Load 5 μL of protein marker for each lane.

Notes:

  1. This product contains a high concentration of glycerol, which can be stored in a liquid state at -20°C and can be used directly. If the temperature of the refrigerator is unstable, resulting in freezing, it can be properly divided and stored at 4°C for at least 6 months.
  2. The separation effect of marker is related to the concentration of PAGE gel. If the concentration of the separation gel is lower than 15%, the bands below 25 kD will not be easily separated, but the green (25 kD) and above bands will not be affected. If the target band is less than 25 kD, it is recommended that the separation gel concentration be greater than or equal to 15%.
  3. This protein molecular weight standard has been denatured at high temperature, please do not heat it at 100℃. 
  4. The effect of membrane transfer is related to the transfer time and the molecular weight of proteins. It is normal if some of the larger molecular weight bands are not successfully transferred to the membrane.
  5. For research use only. Not for use in diagnostic procedures.
