Introduction:

Blue Native PAGE (BN-PAGE) is an electrophoretic technique to resolve protein complexes by molecular weight while retaining their native structure through gel electrophoresis. BN-PAGE can separate proteins and protein complexes in the mass range of 10 kDa to 10 MDa.BN-PAGE substitutes Coomassie brilliant blue G-250 for SDS to make the protein complexes negatively charged and separated in the gel according to the molecular weight of each complex. The samples were dissolved with some mild detergents such as dodecylmaltoside (DM), Triton X-100 and digitonin, etc., so that the complex was separated in a near natural state. Native proteins and complexes migrate as blue bands through BN gels. Protein subunits of complexes can be finally identified by various denaturing techniques such as SDS-PAGE.
